PN HPT® Technology
Polynucleotides are purified DNA fractions used in regenerative and aesthetic medicine to support the biological environment of the skin.
Mastelli's Polynucleotides High Purification Technology—PN HPT®—uses a proprietary, fully controlled purification process to produce highly purified polynucleotides from freshwater-raised salmon trout of European origin.
Conventional dermal fillers are commonly used to restore facial structure or volume. PN HPT® treatments are primarily selected to improve the condition and appearance of the skin.
Depending on the product selected, the treatment may support:
The treatment objective is generally progressive, natural-looking skin improvement rather than an immediate change in facial shape.
PN HPT® has been studied for its interaction with fibroblasts, which contribute to the maintenance of the skin's extracellular matrix.[R3]
Laboratory and clinical research has examined PN HPT® in relation to tissue repair and extracellular matrix organisation.[R3]
Published laboratory research has investigated the scavenging activity of PN HPT®-based hydrogels against reactive oxygen species.[R3]
Clinical studies have reported improvements across selected measures of skin texture, firmness, hydration and radiance.[R2]

Scientific Evidence
Polynucleotides are an active area of research in regenerative and aesthetic medicine.
Published studies involving PN HPT® have examined skin quality, Asian skin rejuvenation, periocular treatment, acne scars, facial rejuvenation and combination treatment with hyaluronic acid. Research outcomes are encouraging, but aesthetic results vary and the overall evidence base continues to develop.
A 2024 study involving 30 Asian subjects evaluated PN HPT® treatment across skin-tone evenness, surface evenness, firmness and glow. The study reported meaningful improvements across several assessed skin-quality categories.
Lim TS et al. Clin Cosmet Investig Dermatol. 2024;17:417–431. [R2]A real-world study evaluated Plinest® 40 mg/2 ml for skin rejuvenation of the face, neck and décolleté and reported improvements across physician- and patient-assessed outcomes.
Real-world PN HPT® assessment. [R4]A prospective randomised study compared highly purified polynucleotides with placebo for moderate-to-severe acne scars and reported improvements in dermal quality and scar appearance.
Araco A, Araco F. Aesthet Surg J. 2021;41(7):866–874. [R5]Published studies have evaluated polynucleotide treatment for periocular skin quality, elasticity and hydration. Not all periocular studies specifically identify Plinest® Eye as the study product.
Palmieri IP et al. Surgical Research. 2023. [R6][R7]A systematic review found encouraging aesthetic applications for polynucleotides while noting variation in study methods and the need for additional high-quality clinical trials.
Systematic review of polynucleotides in aesthetic medicine. [R3]Laboratory and in vivo studies have examined PN HPT® in relation to fibroblast vitality and extracellular matrix organisation, with histological assessments showing tissue repair patterns.

They are not positioned as conventional structural fillers. Their main objective is improvement of skin quality, hydration and elasticity rather than significant facial reshaping or volumisation.
Plinest® contains high-concentration PN HPT® (40 mg/2 ml) for intensive facial bio-regeneration. Newest® combines PN HPT® (20 mg/2 ml) with linear hyaluronic acid (20 mg/2 ml) and mannitol for regeneration together with hydration. Newest® is also indicated for the neck and décolleté.
Plinest® Eye is designed for the delicate periocular area and may help improve skin texture, firmness and elasticity following medical assessment.
Plinest® Eye is primarily a skin-quality treatment. Structural eye bags, prominent fat pads or deep hollowness may require a different treatment approach. A medical consultation is necessary to determine suitability.
Some patients may notice early hydration or freshness. Skin-quality improvements generally develop progressively during and after the treatment course.
The Mastelli guide commonly describes three to four treatments for Plinest®, Plinest® Eye and Newest®. The treating practitioner will personalise the programme.
Temporary raised injection points, redness, swelling, tenderness or bruising may occur. Recovery differs between patients and treatment areas.
Duration varies according to age, skin condition, lifestyle, treatment course and individual biological response. Maintenance treatment may be recommended.
A qualified practitioner may incorporate PN HPT® into a wider treatment plan. Appropriate timing and treatment sequence are important and must be determined by the healthcare professional.
Treatment must be performed by an appropriately qualified and trained healthcare professional, in accordance with local regulations.
No. A medical consultation is required to assess suitability, medical history, allergies, pregnancy or breastfeeding status, current health conditions and other relevant factors.
